چکیده

Several observations have implicated oxidative stress and aggregation of the presynaptic protein-synuclein in the pathogenesis of PD.-Synuclein has been shown to have affinity for unsaturated fatty acids and membranes enriched in PUFAs, which are especially sensitive to oxidation under conditions of oxidative stress. One of the most important products of lipid oxidation is 4-hydroxynonenal (HNE), which has been implicated in the pathogenesis of Parkinson’s disease. Consequently we investigated the effects of the interaction of HNE with-synuclein.

Involvement of 4-hydroxy-2-nonenal Accumulation in Multiple System Atrophy

Recent studies have suggested implications for α-synuclein cytotoxicity in the pathomechanism of multiple system atrophy (MSA). Given in vitro evidence that α-synuclein generates oxidative stress, it is proposed that lipid peroxidation may be accelerated in MSA. To address this issue, we performed an immunohistochemical analysis of protein-bound 4-hydroxy-2-nonenal (P-HNE) in sections of archiv...

A novel lipid hydroperoxide-derived cyclic covalent modification to histone H4.

Previous studies have established that 4-hydroxy-2-nonenal is a lipid hydroperoxide-derived aldehydic bifunctional electrophile that reacts with DNA and proteins. However, it has now been recognized that 4-oxo-2-nonenal is also a major product of lipid hydroperoxide decomposition. Furthermore, 4-oxo-2-nonenal is more reactive than 4-hydroxy-2-nonenal toward the DNA-bases 2'-deoxyguanosine, 2'-d...

Mechanism of destruction of microtubule structures by 4-hydroxy-2-nonenal.

A major end product of lipid peroxidation, 4-hydroxy-2-nonenal (HNE), is an electrophilic alkenal and produces Michael adducts with cellular proteins. It is known that exposure of cultured cells to HNE causes rapid disappearance of microtubule networks. In this study we addressed the mechanism. Immunochemical studies revealed that HNE preferentially modified alpha-tubulin in rat primary neurona...

Heat-solubilized curcumin should be considered in clinical trials for increasing bioavailability.

To the Editor: The recent article by Dhillon et al. (1) is of great interest to those working with curcumin. As the investigators suggest, the usefulness of curcumin could be minimized because of its poor oral bioavailability (2). Data from this study show that only 22 to 41 ng/mL were detectable in plasma even when 8 g curcumin/day was given orally (1). Curcumin levels in the microgram range h...

Modification of heat shock protein 90 by 4-hydroxynonenal in a rat model of chronic alcoholic liver disease.

Lipid peroxidation during oxidative stress leads to increased concentrations of thiol-reactive alpha,beta-unsaturated aldehyde, including 4-hydroxy-2-nonenal (4-HNE) and 4-oxo-2-nonenal (4-ONE). These aldehydes have a documented ability to disrupt protein function following adduct formation with specific residues. Therefore, to identify 4-HNE-modified proteins in a model of ethanol-induced oxid...
